چکیده

An enzyme preparation obtained from bovine lymphosarcoma tissue catalyzes the DNA-dependent synthesis of RNA. Native DNA is a more efficient primer in the reaction than heat-denatured DNA.

The role of template-primer in protection of reverse transcriptase from thermal inactivation.

We compared the thermal stabilities of wild-type recombinant avian myeloblastosis virus (AMV) and Moloney murine leukemia virus (M-MLV) reverse transcriptase (RT) with those of mutants of the recombinant enzymes lacking RNase H activity. They differed in resistance to thermal inactivation at elevated temperatures in the presence of an RNA/DNA template-primer. RNase H-minus RTs retained the abil...

Protection of avian myeloblastosis virus (AMV) DNA polymerase by substrates against heat inactivation.

Purified DNA polymerase from AMV was exposed to thermal inactivation in the presence and absence of various compounds required for maximal activity. A synergistic protective effect was noted when all the components were present. The enzyme showed a distinct preference for primed templates, especially for its native 70S RNA. Heat inactivation studies also suggested that the active sites for RNas...

Optimization of RNA Extraction from Rat Pancreatic Tissue

Background: Optimized RNA extraction from tissues and cell lines consists of four main stages regardless of the method of extraction: 1) homogenizing, 2) effective denaturation of proteins from RNA, 3) inactivation of ribonuclease, and 4) removal of any DNA, protein, and carbohydrate contamination. Isolation of undamaged intact RNA is challenging when the related tissue contains high levels of ...

P-207: Deltamethrin-Induced Derangements Correlates with Estrogen Synthesis, Angiogenesis and Oxidative Stress

Background: Deltamethrin (DTM) is a pyrethroid derivative, which is the most potent products of Type 2 insecticides and acts by delaying the closure of sodium channels. Estrogen as a vascular growth factor on ovarian tissue plays an essential impact further to its involvement in follicular growth. On the other hand, the role of angiogenesis on ovarian tissue in inhibiting the apoptotic provoker...

Novel mutations in Moloney Murine Leukemia Virus reverse transcriptase increase thermostability through tighter binding to template-primer

In an effort to increase the thermostability of Moloney Murine Leukemia Virus reverse transcriptase (MMLV RT), we screened random and site-saturation libraries for variants that show increased resistance to thermal inactivation. We discovered five mutations E69K, E302R, W313F, L435G and N454K that collectively increase the half-life of MMLV RT at 55 degrees C from less than 5 min to approximate...
